In particular, the observation of conformational changes within these biomolecules or during interactions have become directly accessible. Here, I will present how we develop and apply single-molecule techniques to characterize molecular machines. I will introduce a combination of magnetic tweezers and single-molecule FRET to observe two coordinates simultaneously.
